Yes. A registered trademark (or a pending trademark application with a serial number) in an Amazon-recognized jurisdiction is a core requirement for enrolling in Amazon Brand Registry.

A pending application is sufficient; full registration is not required. Amazon accepts the serial number issued by the IP office at the time of filing. If the application is ultimately refused, Amazon may revoke Brand Registry access.
